TCID₅₀ Assay

Overview
A gold-standard assay to determine the dilution of a virus that infects 50% of cell cultures, widely used for virology research and vaccine potency evaluation.

Principle

  • Serial dilutions of virus are added to cells in 96-well plates.

  • After incubation, cytopathic effects (CPE) are observed.

  • Viral titer is calculated using the Reed–Muench method.

Applications

  • Standardizing viral inoculum for experiments

  • Vaccine potency and challenge studies

  • Antiviral drug efficacy evaluation
